Phospholipids are the main constituents of the cellular membranes in all organisms, ranging from single-celled archaea to highly complex plants and mammals. According to conventional wisdom, the chemical backbone of phospholipids in archaea is 'right-handed', but left-handed in all other organisms. The little-understood mammalian phospholipid bis(monoacylglycero)phosphate (BMP), however, is a possible exception to this rule. Peter Greimel, HuiHui Tan and their colleagues at the RIKEN Advanced Science Institute, Wako, have now obtained the first proof that BMP is indeed right-handed.
BMP is found only in mammals, and is a common -- but minor -- constituent of all animal tissues. The internal membranes of the waste treatment and recycling centers of cells -- so-called late endosomes and lysosomes -- have a higher proportion of BMP than any other animal membranes. This raised an intriguing question, explains Greimel: "How does BMP escape degradation inside these organelles, unlike all the other lipids and proteins?" This led to the [hypothesis] that BMP might actually be right-handed, allowing it to avoid attack by the organelle's digestive enzymes that are only capable of recognizing, and therefore destroying, left-handed lipids. All previous attempts to confirm the suspected unusual handedness -- or chirality -- of BMP had hit problems.
The research team first synthesized all the possible variations of BMP. They then reacted these variations with the chiral shift reagent D-camphor. "The D-camphor induced a change in the spectroscopic behavior of each synthetic BMP analogue," explains Greimel. This meant that the nuclear magnetic resonance (NMR) -- a common spectroscopic technique -- spectra of the BMP analogues were different enough to be distinguished from each other.
Next, the researchers isolated natural BMP from baby hamster kidney cells using standard techniques, then reacted it with D-camphor under very gentle conditions and analyzed it spectroscopically. They then compared the NMR spectra of the natural BMP derivative and the synthetic molecules. "Analysis of the spectroscopic data revealed that natural BMP is exclusively right-handed," Greimel says.
"Since BMP is right-handed, it means it most likely originated from the same common ancestor as archaea," he explains. The research team now plans to identify the enzymes involved in the biosynthesis of BMP, with the hope that detailed knowledge of this biosynthetic pathway will eventually lead to a better understanding of how life evolved on Earth.
Additionally, Greimel says that "now we know the [chirality] of the molecule, we can think about synthesizing analogues in order to develop novel drugs, in this case to treat lysosomal storage diseases."

One hundred US special forces trainers are working with the Ugandan military to put an end to the rebel group. And while they may have succeeded in sending the group on the run, the LRA has proven dangerous in its desperation.
The latest attacks have occurred in the last 30 days, with LRA attacks reported in the village of Bagulupa, 35 miles east of Dungu in the eastern region of the Democratic Republic of Congo. The attacks occurred on Feb. 10 and 24, and appear to have been standard raids for food. One person was killed, and 17 villagers abducted, probably for use as porters or sex slaves, according to the Office of the United Nations High Commissioner for Refugees. Most of the villagers of Bagulupa have fled toward the larger town of Dungu, the UNHCR says.
Fatoumata Lejeune-Kaba, the UNHCR?s spokeswoman in Geneva, voiced concern about the recent uptick in violence in the DRC, after a six-month lull in the latter part of 2011.
?UNHCR is very concerned at the recent displacement of several thousand people as a result of renewed attacks by the Lord's Resistance Army (LRA), in the Democratic Republic of the Congo's Orientale province,? said Ms. Lejeune-Kaba, adding that there have been 20 such attacks since the beginning of the year.
As suggested by its name, the Lord?s Resistance Army is a quasi-religious militia founded in 1987 in northern Uganda by Joseph Kony. The group originally fought in defense of what it believed to be the grievances of the northern Ugandan people, but its mission shifted toward one of terror and survival, and it has remained on the run ever since. LRA raids have struck deep into South Sudan, Uganda, the Central African Republic and the Democratic Republic of Congo. In Congo?s Orientale Province, an estimated 320,000 people have been displaced from their homes?and 30,000 more Congolese refugees have fled into neighboring Central African Republic and South Sudan.
Having been invaded in the past by Ugandan and Rwandan troops in the late 1990s, in their successful bid to topple the Congolese government of the day, the government of the Democratic Republic of Congo refuses to allow Ugandan troops to enter Congolese territory in pursuit of the LRA. But regional security experts say the Congolese troops in Orientale Province are ?notoriously incapable of dealing with the rebels.?
The US government, which has sent 100 special operations forces commandos to help train the Ugandan Defense Forces who are hunting down Kony and the LRA, says that sustained pressure from the militaries of the CAR, Uganda, South Sudan, and the DRC have forced the LRA to go on the run.
"With our support, these four military forces continue to make progress in reducing the LRA numbers and keeping them from regrouping. We believe it is critical the militaries in the region continue to work together to keep the pressure on the LRA and protect their own citizens. As we have seen in the past, the LRA will exploit any reduction in military or diplomatic pressure to regroup and rebuild their forces," Karl Wycoff, the US deputy assistant secretary for African affairs, in a telephone briefing on 22 February, told IRIN news agency.
PARIS (Reuters) - France's Foreign Minister said on Wednesday he was skeptical renewed talks between six world powers and Iran would succeed as Tehran was still not sincere in its willingness to negotiate over the future of its nuclear program.
EU foreign policy chief Catherine Ashton, who represents the United States, Russia, China, France, Britain and Germany in dealings with Iran, said on Tuesday the six powers had accepted an Iranian offer for talks on its disputed nuclear program.
"I am a little skeptical ... I think Iran continues to be two-faced," French Foreign Minister Alain Juppe told France's i-Tele television.
"That's why I think we have to continue to be extremely firm on sanctions, which in my view are the best way to prevent a military option that would have unforeseeable consequences."
The Islamic Republic's approach to the six powers, in a letter dated February 14, comes as it suffers unprecedented economic pain from expanding sanctions against its oil and financial sectors.
Western states are likely to tread cautiously in talks, mindful of past accusations that Iran's willingness to negotiate has been a stalling tactic to blunt pressure rather than a genuine effort to reach agreement.
With Israel speaking increasingly loudly of resorting to military action, the talks could provide some respite in a crisis that has driven up oil prices and threatened to suck the United States into its third major war in a decade.
French President Nicolas Sarkozy was the first among leaders of the six powers to push for tighter sanctions on Iranian oil and finance, however Juppe on Wednesday signaled that France was wary of resorting to a military strike against Iran.
"There is still a debate in Israel (about a military strike) and it's our responsibility to bring to Israel's attention the unforeseeable consequences it would have," he said.
Sarkozy said in January that time was running out for efforts to avoid military intervention in Iran.
