New cases of rare genetic disorder identified

Scientists at the University of Liverpool, working with international partners, have shown a rare genetic disease, that causes crippling osteoarthritis in the spine and major joints, is far more prevalent worldwide than previously thought.

Researchers have identified more than 100 new cases of the disease, called alkaptonuria (AKU) in a small community in Vellore, India, bringing the total number of patients there to 130. Approximately 40 patients have been found in a village in Southern Jordan, but previously the disease had been unknown to healthcare workers in the region. There were only four known cases in the UK in 2003, but this has since risen to more than 80 patients across the country.

Research findings suggest that more work is needed to raise the profile of the disease in countries around the world so that it can be correctly identified by medics. It is thought to affect one in 250,000 people worldwide, but many remain undiagnosed or misdiagnosed as osteoarthritis.

Scientists at Liverpool are bringing international researchers together as part of the AKU Society to establish a global network that will help further understanding of the condition within communities and healthcare practices. The AKU Society was established in Liverpool in 2003 and supports patients diagnosed with the disease.

Patients being treated for AKU do not have enough of the enzyme, homogentisic acid oxidase, which causes acid to build up in the body. Some of this acid is eliminated in the urine, but the remainder is deposited in body tissue where it is toxic. As a result, a black pigment, called ochronosis, forms and binds to bone, cartilage and skin. This can cause erosion of the cartilage and patients often have to undergo joint replacement surgery.

Dr Lakshminarayan Ranganath, researcher at the University's Institute of Ageing and Chronic Disease and clinician at the Royal Liverpool University Hospital, said: "AKU was the first genetic disease to be identified by scientists. It was described in 1902 by an English physician, but more than 100 years later there is still no cure for the disease. A drug candidate has recently been identified and is awaiting clinical trials to see if it could be used to treat AKU patients. We want to identify as many people with the condition as possible to ensure these important trials go ahead.

"Recent findings in India and Jordan show us that this disease is more prevalent than current statistics suggest and we urgently need to get research out into healthcare practices around the world so that cases can be identified and diagnosed correctly. We want to expand our international links as well as promote PhD opportunities to young researchers looking to progress our knowledge of the disease.

Professor James Gallagher, AKU researcher at the University's Institute of Ageing and Chronic Disease, said: "We are currently working to understand why the black pigment that attaches itself to joint cartilage only attaches to certain areas, whilst other sections of cartilage remain pigment-free. If we can understand the mechanisms of how the pigment binds in the body then we may be able to prevent the disease developing."

Isaac Jebaraj, Professor of Orthopaedic Surgery at the Christian Medical College, Vellore, India, said: "My work is focused on a Romani Community in South India, where we have identified approximately 100 cases of AKU. It is important for us to reach these communities as traditionally they do not attend hospitals for aliments, preferring to use herbs and oils instead. As such we anticipate that there are many more people in India with this condition that have yet to be diagnosed with the condition.

"We have now treated many of the community with anti-inflammatory drugs and some have had surgery for joint replacement. To ensure that potential new treatments reach these communities, it is important that scientists work together to progress with drug trials."

Little-known brain disease rips apart lives of victim, loved ones

More than a year after the diagnosis, Maureen Bryant had grown accustomed to making excuses for her husband.

When Stu stood behind a tattooed woman in line at Panda Express, and said loudly, "Wow, that's a lot of tattoos," Moe stepped between him and the woman and apologized.

When he repeatedly wandered into the house that was being built down the street ? despite the "No Trespassing" sign and the fence ? she explained to the owner that he was just curious.

Possibly the most embarrassing episode occurred when they were coming home from dinner, and she dashed into a mini-mart at a marina in Oxnard to buy milk.

"Do you know that man?" the owner said, seeing Stu in the car. "He's been stealing from me."

Moe remembers being baffled.

Muffins and cheesecake, the owner said, showing her photos from the store's video camera. "But I can't catch him. He drives away in the golf cart too fast."

Moe offered to pay for what Stu had taken. "He has no control over what he does," she said to police when they arrived.

Sometimes she will say that he has a brain tumor. It's easier than saying frontotemporal dementia and having to describe how the disease has stripped away his self-restraint. Stu is no longer aware of how to behave in the company of others, and like an unknowing child, will blurt out the first thing that comes to mind or will act without considering the consequences.

Moe realizes that this is difficult to understand. Being rude or disrespectful seems like a deliberate choice, and Stu, 60, has a quick smile and an easy laugh. It's easy for people to write him off as inconsiderate and self-centered.

This is what Moe and her daughters, Katy and Jessica, did before they discovered how sick he was.

Once called Pick's disease and now FTD, frontotemporal dementia hides its symptoms well. Like Alzheimer's, it is the result of the degeneration of neurons caused by an abnormal accumulation of proteins in the brain.

In early stages, Alzheimer's affects predominantly the hippocampus in the inner part of the brain, where memories are formed, and FTD affects the frontal and temporal lobes, above and behind the eyes, where social emotions are formed. Alzheimer's patients sense a fuzziness in their thinking ? a word misplaced, a confusion between the past and present ? and realize their errors. Most patients with FTD have no such insight into their disease.

Jessica, 22, first noticed her father becoming more selfish and temperamental when they lived in Sacramento. He doted upon his daughters and would do anything for them, but then that seemed to stop. Where once he let Jessica choose what to watch on TV, now he held on to the remote and got angry if she changed the channel.

The tragedy of FTD, according to Mario Mendez, the UCLA neurologist treating Stu, is its elusive diagnosis. The presence of the disease can be confirmed only through an autopsy, and Mendez reached his conclusion about Stu based on specific behaviors and brain scans.

The disease begins with a change in personality that's often seen as volitional. Introverts become extroverts. Once-agreeable employees act rude. Formerly faithful spouses have affairs, and while they acknowledge their transgressions, they are unable to appreciate the implications.

Some end up institutionalized because their behavior looks like mental illness. Some are incarcerated when they break the law. A diagnosis of FTD led to reduced charges against a New York City physician who cut his initials into the stomach of a patient, and what could have been as many as 25 years in prison ended up as five years' probation.

Researchers believe that between 30,000 and 50,000 people in the United States have FTD. There is no cure, and the disease leads to death, often within eight years, either from accidents or secondary infections.

Dow approaches 13,000, and maybe a record to come

(AP) ? It was just last summer that the Dow Jones industrial average shed 2,000 points in three terrifying weeks. Investors had a host of things to worry about, including the possibility of another recession.

Now the Dow is within reach of the rarefied 13,000 mark ? a level it hasn't seen since May 2008, four months before the financial system almost came apart.

A strong one-day rally ? caused by a deal on bailout money for Greece, perhaps, or an unexpectedly positive economic report ? could put it over the top.

What's more, the average is just a 10 percent rally from an all-time high. And 10 percent rallies can happen fast these days.

The stomach-turning summer is a bad memory. Europe appears to be getting its act together, last summer's downgrade of the U.S.' credit rating was quickly forgotten, Washington is mostly behaving, and recession fears are gone.

"There are signs that the economy is getting back on its feet and the market is reacting to that," says John Prestbo, executive director of Dow Jones Indexes. "The mood is just better in this country than it has been for a while."

On Wall Street, too. The Dow traded Tuesday at 12,878, a 21 percent rally from Oct. 3, its low point for last year. In January, the average rose more or less in a straight line and added 3.4 percent, its best start to a year since 1997.

From here, the record is tantalizingly close ? 14,164.53, reached Oct. 9, 2007, when the investment houses Bear Stearns and Lehman Brothers still existed and the unemployment rate was 4.7 percent.

A 10 percent surge may seem like a lot, but it's really not. The Dow has gained almost 15 percent since Nov. 25, just 10 weeks ago.

Though there's a long way to go to get the country back to economic health, there are pockets of encouragement. Unemployment is still 8.3 percent, but it's the lowest since February 2009. Economic output grew every quarter last year.

Corporate earnings growth has slowed, but analysts think it will pick up again later this year. Investors, always wary of uncertainty, may even be encouraged by some clarity in the Republican presidential nominating race.

Investors are no longer just trying to stem their losses, says Mark Lehmann, president of JMP Securities in San Francisco: "They're playing a little offense. Six months ago, they were playing defense."

There's evidence that the rally has room to run. In a popular measure of how expensive stocks are, the 30 companies that make up the Dow are trading at an average of about 13 times their annual earnings per share.

The last time the Dow was at 13,000, in May 2008, stocks were trading for about 15 times earnings. Stock-market research firm Birinyi Associates estimates Dow stocks have traded at an average of 16 times earnings over the past two decades.

The fire-sale discounts have already come and gone, though. Those were back in early 2009, when the Dow bottomed at 6,547.05, its Great Recession low ? a little more than half the level now. Back then, Dow stocks traded at nine times earnings.

Not everyone believes the rally will last. Joe Gordon, managing partner at Gordon Asset Management in North Carolina, is dubious. He cites the unresolved European debt crisis, the U.S.' historically high national debt and the millions of people who have given up looking for work, part of the so-called underemployed.

"This is like drinking a lot of coffee in the afternoon," says Gordon. "It perks you up, then once it fades 45 minutes later you're even more tired."

Another wrinkle is that the Dow tracks just 30 companies, so it doesn't take the full pulse of the market. The Standard & Poor's 500, with its much larger roster, is still 16 percent away from its all-time high.

"It's 30 stocks," says Rob Leiphart, an analyst at Birinyi. "It doesn't give you a representation of anything."

But despite its size, the Dow is the market gauge that penetrates the public consciousness, generating headlines and water cooler buzz more than the less publicized S&P.

That's important because the stock market, even if it has no direct bearing on the fundamentals of the economy, is a psychological motivator of spending because of something known as the wealth effect.

Even people with no stock investments will let their decisions be influenced by swings in the Dow. When it's up, we tend to feel richer and spend more. When it's down ? think back to the 500-point daily declines of 2008 ? we tend to feel poorer and spend less.

There's good reason the Dow has pull over the financial mood of the country. Its 30 stocks account for 25 to 30 percent of the market value of all U.S. public companies, and about 40 percent of the dividends, Dow Jones Indexes estimates.

"Nothing of substance can happen in this economy without these companies feeling it," Prestbo says.

A handful of companies have an outsized impact on the index. The Dow is a price-weighted average, which means companies with more expensive stocks have more power to drive the average higher or lower.

If you invest $30 in a mutual fund tracking the Dow, you don't have a dollar riding on each company. Four times as much of your money would end up on Home Depot, which is trading around $45, than Alcoa, trading around $11.

IBM, the highest-priced stock in the Dow, had a giant influence last year. The Dow rose 5.5 percent in 2011, but without IBM it would have risen only 3.4 percent, according to Leiphart's calculations.

If you were to cut out the next three stocks on the list, McDonald's, Chevron and ExxonMobil, then the Dow would have finished down 0.25 percent for the year.

The flip side is that stocks like Chevron, Exxon Mobil, Microsoft and Intel trade well below the 13 times earnings for the full Dow. If they catch up, it could be enough to power the average to a record.

"Night of the Living Dead" actor Bill Hinzman dies (Reuters)

LOS ANGELES (Reuters) ? Bill Hinzman, who was cast on-the-spot as a zombie in 1968 cult film "Night of the Living Dead" in a role that earned him the longlasting admiration of horror fans, has died at age 75, his daughter said.

Hinzman died of cancer on Sunday evening at his home in Darlington, Pennsylvania, his daughter Heidi Hinzman told Reuters.

He played a prominent role in Director George Romero's low-budget, black-and-white 1968 film "Night of the Living Dead," which is credited with revolutionizing the zombie genre.

Hinzman was working as an assistant cameraman on the film when Romero decided to cast him as the zombie in the movie's opening sequence.

"We'd like to tell the story that it was a hard audition session, but Bill was there and old enough and thin enough and he had an old suit," said the film's producer Russ Streiner.

Dressed in that suit, Hinzman appears at a Pennsylvania cemetery and attacks a pair of young siblings by knocking the man's head against a tombstone and chasing the woman.

He lurches across an open field and eventually chases the woman into a farmhouse.

Fans called Heinzman "No. 1 zombie," said Streiner, who also played a character named Johnny who fought with Hinzman in the cemetery.

"He was a very popular guy, he was very accessible to the fans," Streiner added.

Hinzman also went on to direct and star in the 1980s horror movies "The Majorettes" and "FleshEater."

He will be cremated, as he had asked, said Heidi Hinzman.

"He always joked with me that if he got buried he would come back," she said.

Court unseals indictment of Native American gang members (Reuters)

MINNEAPOLIS (Reuters) ? Federal authorities were looking for three members of a Native American gang on Wednesday charged alongside 21 others in a 47-count federal indictment with racketeering and drug offenses.

Most of the indicted "Native Mob" gang members were already in custody after a January indictment in a case prosecutors hoped would deal a blow to a gang that formed in Minneapolis in the 1990s and has about 200 members across the upper Midwest.

Members have preserved "the power, territory, reputation and profits of the enterprise through the use of intimidation and violence, including murders, attempted murders, assaults, robberies and threats of violence," the indictment said.

The wide-ranging indictment alleges several attempted murders of members of rival gangs, robberies, assaults, threats against witnesses cooperating with law enforcement and numerous shots fired into houses from 2003 through 2011.

In one incident, two gang members were accused of shooting a witness three times while he was holding his 5-year-old daughter in an attempted murder. In another, a gang member tossed scalding water into the face of a person protecting another victim who was cooperating with authorities.

Gang members sold or were arrested for possessing various illegal drugs including heroin, cocaine, crack, ecstasy and methamphetamine in the years covered by the indictment.

Members of the gang often wear red and black clothing and have tattoos with the gang name or initials, sometimes incorporating a medicine wheel, bear paw or other symbols, the indictment said.

The indictment, ordered unsealed on Tuesday, outlines a gang structure aimed at establishing a racketeering charge, including monthly meetings and regional gang representatives.

It said that top leaders maintained discipline, others took care of money questions and provided cash to members serving prison sentences, and lower level leaders responded to external threats or punished members who broke rules.

Markets buoyant ahead of run of US data (AP)

LONDON ? Solid manufacturing surveys from around the world helped boost markets on Wednesday as attention turned towards the state of the U.S. economy in the run-up to a crucial jobs report.

With figures showing that China's manufacturing sector is growing solidly and Europe's performing better than forecast, investors are hopeful that equivalent U.S. figures later will show the world's largest economy is recovering.

Investors will then focus on the monthly hiring figures from private payrolls agency ADP, which will help shape expectations of Friday's official jobs data, non-farm payrolls. The most closely-watched indicator of growth in the U.S., the payrolls figures often set the market tone for a week or two after their release. The expectation is currently that the U.S. created another 180,000 jobs in January.

"Last month the ADP figure caught everyone out, smashing through expectations," said Chris Beauchamp, market analyst at IG Index. "Today's forecast is for a gain of 185,000, and this will need to be met if today's rally is not to prove as short-lived as yesterday's.".

Ahead of the ADP survey, which will be released an hour before the bell on Wall Street, investors were optimistic.

In Europe, the FTSE 100 index of leading British shares was up 1.5 percent at 5,766 while Germany's DAX rose 2.1 percent to 6,593. The CAC-40 in France was 1.7 percent higher at 3,353.

Wall Street futures also pointed firmly higher ? Dow futures were up 0.7 percent at 12,670 while the broader Standard & Poor's 500 futures rose by the same rate to 1,318.

The focus on the U.S. will prove a welcome diversion for some traders from monitoring the daily developments in Europe's debt crisis.

There are signs that the crisis has eased, for now. EU leaders have agreed this week to push ahead with a closer fiscal union and borrowing rates for Italy and Spain are down sharply from just a couple of months ago, suggesting increased investor confidence.

Much hinges on Greece, where the outlook also appeared brighter. Hopes were growing that a debt-reduction deal between the country and its private creditors will be concluded soon alongside a second bailout from the eurozone and the International Monetary Fund.

"We still await news from the Greek debt restructuring talks although the few comments that have been made over the last 48 hours have suggested that progress is being made," said Gary Jenkins, managing director at Swordfish Research.

The sense of an easing in Europe's debt woes helped stocks enjoy a stellar start to the year, with many markets recovering a large chunk of their late-2011 losses. Overall, U.S. shares had their best start in 15 years.

The improving backdrop has helped shore up the euro, too, which was trading 0.3 percent higher at $1.3127.

Earlier in Asia, stock markets lacked the same momentum seen in Europe.

Tokyo's Nikkei 225 edged up less than 0.1 percent to close at 8,809.79 but Hong Kong's Hang Seng ended down 0.3 percent to 20,333.37. Mainland China's main index in Shanghai also fell 1.2 percent to 2,268.08.

Benchmark oil for March delivery gained 72 cents to $99.20 per barrel in electronic trading on the New York Mercantile Exchange. The contract fell 30 cents to end at $98.48 per barrel in New York on Tuesday.

Scientists intrigued by atoms from beyond the solar system

NASA's IBEX spacecraft has snagged atoms that came from outside our solar system. Interstellar space includes the raw material that becomes new stars, planets, and organic life.?

A NASA satellite orbiting Earth has captured "aliens" from interstellar space ? but with names like hydrogen, helium, oxygen, and neon, these are outsiders are actually quite welcome.

By snagging these different elements at the same time, NASA?s Interstellar Boundary Explorer (IBEX) is for the first time allowing astronomers to compare how elements are distributed in interstellar space versus within our own solar system.

The composition of interstellar space is of particular interest to scientists because stars seed it with a range of elements ? including nitrogen, oxygen, iron ? when they end their lives. Once part of the interstellar medium, these elements become the raw material available for recycling into new stars, as well as planets, plants, and animals.

The new data are already helping scientists gain a fuller understanding of the corner of the Milky Way in which our solar system currently finds itself.

IBEX is designed to study the boundary between interstellar space and the heliosphere ? the region of space influenced by the charged particles of the solar wind and the sun's magnetic field. Cosmic radiation, which consists of charged particles, is mostly deflected by the heliosphere. But neutral atoms, which carry no electrical charge, can penetrate the sun?s protective shield. Tuesday?s IBEX report focuses on these neutral atoms.

IBEX's sampling of neutral atoms has revealed intriguing differences in composition between the material in the solar system and in the interstellar cloud the solar system is passing through. IBEX found that for every 20 neon atoms reaching its detectors from the interstellar medium, it also was detecting 74 oxygen atoms. The solar system is richer in oxygen, with 111 oxygen atoms for each 20 neon atoms.

That points to two possibilities, says David McComas, a researcher at the Southwest Research Institute in San Antonio, and the mission's lead scientist. Either the sun formed in a different part of the interstellar cloud it now occupies, where oxygen was more plentiful, or the missing oxygen is cradled in dust and ice in the cloud.

If the differences are intriguing now, wait another 100 to 3,000 years or so. That's when the solar system is expected to exit the cloud it's now crossing into far different surroundings, the IBEX researchers say.

In its current surroundings, the solar system is encountering a weaker interstellar headwind than previous estimates suggested, the IBEX team reports. Measurements from the European Space Agency's now-defunct Ulysses solar observatory indicated that helium was entering the solar system at 59,000 miles an hour. IBEX's more-precise measurements put the speed at 52,000 miles an hour.

This reduction in speed means that the heliosphere sports a different shape than previously estimated, notes Dr. McComas. Currently, that is most important to modelers trying to map the various forces at play in the solar system.

?Frankly, all the modelers now have to go back and try to get their models to work with a very different balance? between the pressure the heliosphere is encountering from the headwind and the interplay between the heliosphere's magnetic field and the galactic magnetic field, he says. Both affect the shape of the heliosphere.

Understanding the shape of the heliosphere could be more broadly important if the solar system heads into quarters of the galaxy with much greater headwinds. In principle, those headwinds could push the leading edge of the heliosphere closer to the sun ? to the point that some planets could spend part of their orbit outside the sun?s protective envelope. ?

The IBEX results appear in a special February supplement to the Astrophysical Journal.
